Description

Description: This assay employs a two-site sandwich ELISA to quantitate IKZF3 in samples. An antibody specific for IKZF3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IKZF3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IKZF3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IKZF3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: IKZF3 encodes a member of the Ikaros family of zinc-finger proteins. Three members of this protein family (Ikaros, Aiolos and Helios) are hematopoietic-specific transcription factors involved in the regulation of lymphocyte development. This gene product is a transcription factor that is important in the regulation of B lymphocyte proliferation and differentiation. Both Ikaros and Aiolos can participate in chromatin remodeling.
Regulation of gene expression in B lymphocytes by Aiolos is complex as it appears to require the sequential formation of Ikaros homodimers, Ikaros/Aiolos heterodimers, and Aiolos homodimers. At least six alternative transcripts encoding different isoforms have been described
